The change in phenols, polysaccharides and volatile profiles of noni juice from laboratory- and factory-scale fermentation was analyzed during a 63-day fermentation process. The phenol and polysaccharide contents and aroma characteristics clearly changed according to fermentation scale and time conditions. The flavonoid content in noni juice gradually increased with fermentation. Seventy-three volatile compounds were identified by solid-phase microextraction coupled with gas chromatography–mass spectrometry (SPME-GC-MS). Methyl hexanoate, 3-methyl-3-buten-1-ol, octanoic acid, hexanoic acid and 2-heptanone were found to be the main aroma components of fresh and fermented noni juice. A decrease in octanoic acid and hexanoic acid contents resulted in the less pungent aroma in noni juice from factory-scale fermentation. The results of principal component analysis of the electronic nose suggested that the difference in nitrogen oxide, alkanes, alcohols, and aromatic and sulfur compounds, contributed to the discrimination of noni juice from different fermentation times and scales. 相似文献

Abnormally increased resorption contributes to bone degenerative diseases such as Paget’s disease of bone (PDB) through unclear mechanisms. Recently, the optineurin (OPTN) gene has been implicated in PDB, and global OPTN knockout mice (Optn−/−) were shown to exhibit increased formation of osteoclasts (osteoclastogenesis). Growing evidence, including our own, has demonstrated that intracellular reactive oxygen species (ROS) stimulated by receptor activator of nuclear factor kappa-B ligand (RANKL) can act as signaling molecules to promote osteoclastogenesis. Here, we report that OPTN interacts with nuclear factor erythroid-derived factor 2-related factor 2 (NRF2), the master regulator of the antioxidant response, defining a pathway through which RANKL-induced ROS could be regulated for osteoclastogenesis. In this study, monocytes from Optn−/− and wild-type (Optn+/+) mice were utilized to differentiate into osteoclasts, and both qRT-PCR and tartrate-resistant acid phosphatase (TRAP) staining showed that the Optn−/− monocytes exhibited enhanced osteoclastogenesis compared to the Optn+/+ cells. CellROX® staining, qRT-PCR, and Western blotting indicated that OPTN deficiency reduced the basal expression of Nrf2, inhibited the expression of NRF2-responsive antioxidants, and increased basal and RANKL-induced intracellular ROS levels, leading to enhanced osteoclastogenesis. Coimmunoprecipitation (co-IP) showed direct interaction, and immunofluorescence staining showed perinuclear colocalization of the OPTN-NRF2 granular structures during differentiation. Finally, curcumin and the other NRF2 activators attenuated the hyperactive osteoclastogenesis induced by OPTN deficiency. Collectively, our findings reveal a novel OPTN-mediated mechanism for regulating the NRF2-mediated antioxidant response in osteoclasts and extend the therapeutic potential of OPTN in the aging process resulting from ROS-triggered oxidative stress, which is associated with PDB and many other degenerative diseases.Subject terms: Mechanisms of disease, Stress signalling相似文献

6–8 mass% Y2O3 stabilized ZrO2 (6–8YSZ) thermal barrier coatings (TBCs) are widely applied to protect the hot ends of gas turbines in large navy ships. In this work, the 8YSZ TBCs were prepared by air plasma spraying technique, and their microstructure and phase composition were investigated. The hot corrosion mechanism of YSZ TBCs in molten salts consisting of 80% Na2SO4?+?20% NaCl at 900 °C was comprehensively analyzed. The results showed that the corrosion product Y2(SO4)3 was formed due to the reaction between Na2SO4 media and the stabilizer Y2O3. As the result of the depletion of Y2O3 phase, the transformation from the tetragonal phase to monoclinic phase of ZrO2 could not been totally inhibited, which consequently induced the 4–6 vol.% expansion and more cracks of YSZ TBCs. Meanwhile, the cracks could work as transfer paths for oxygen and molten salts. The kinetic analysis on hot corrosion also showed that more reaction products (from 2 to 8.1 mg cm?1) were generated from 20 to 60 h due to more cracks generated by molten salts and oxygen infiltrating. More thermal grown oxides generated between ceramic layer, bonding layer and substrate, and the volume expansion caused by phase transition, increased the stresses in the coatings. Consequently, the peeling-off failure of 8YSZ TBCs could happen.
The horizontal convection in a square enclosure driven by a linear temperature profile along the bottom boundary is investigated numerically by using a finite difference method. The Prandtl number is fixed at 4.38, and the Rayleigh number Ra ranges from107 to 1011. The convective flow is steady at a relatively low Rayleigh number, and no thermal plume is observed, whereas it transits to be unsteady when the Rayleigh number increases beyond the critical value. The scaling law for the Nusselt number Nu changes from Rossby's scaling Nu ~ Ra~(1/5) in a steady regime to Nu ~ Ra~(1/4) in an unsteady regime, which agrees well with the theoretically predicted results. Accordingly,the Reynolds number Re scaling varies from Re ~ Ra~(3/11) to Re ~ Ra~(2/5). The investigation on the mean flows shows that the thermal and kinetic boundary layer thickness and the mean temperature in the bulk zone decrease with the increasing Ra. The intensity of fluctuating velocity increases with the increasing Ra. 相似文献
The effects of pre-equilibrium emission and secondary decay on the determination of the freeze-out volume are investigated using the isospin-dependent quantum molecular dynamics model accompanied by the statistical decay model GEMINI. Small-mass projectiles and large-mass targets with central collisions are studied at intermediate energies. It is revealed that the proton yields of pre-equilibrium emission are smaller than those of secondary decay. However, the determination of the freeze-out volume from the proton yields is more easily affected by pre-equilibrium emission. Moreover, the percentage of proton yields in the freeze-out stage is found to be approximately 50%. 相似文献
